Ene-reductase is a class of oxidoreductase capable of catalyzing the reduction of activated carbon-carbon double bonds. It is mainly used to selectively reduce the C=C double bonds in substrates such as enone, enal, enoic acid, enoate, and nitroalkene into saturated structures. Compared with traditional metal-catalyzed hydrogenation or chemical reduction processes, ene-reductase operates under milder reaction conditions, typically in aqueous systems or mild organic/aqueous systems, and offers high regioselectivity, chemoselectivity, and enantioselectivity, making it particularly suitable for chiral center construction and the synthesis of high-value intermediates.

The upstream of ene-reductase mainly includes microbial strain resources, enzyme gene sequences, expression vectors, fermentation media, cofactor recycling systems, immobilization carriers, and enzyme formulation excipients. The midstream involves enzyme screening, directed evolution, fermentation expression, purification, immobilization treatment, and process scale-up. Downstream applications are mainly concentrated in pharmaceutical biocatalysis, agrochemical intermediate, flavor and fragrance, and specialty fine chemical, where ene-reductase is used for selective reduction of unsaturated substrates, chiral center construction, and green synthesis process development.
