Compared with transaminase and imine reductase, amine dehydrogenase can directly use ammonia or ammonium salts as the nitrogen source, converting carbonyl compounds into chiral amines or amine-containing intermediates in a single step, with relatively simple by-products and high atom economy. Due to its mild reaction conditions, good selectivity, and low risk of metal residue, amine dehydrogenase has strong application value in the green synthesis of chiral amines, API intermediates, and high-value fine chemicals.

The upstream of amine dehydrogenase mainly includes microbial strain resources, enzyme gene sequences, expression vectors, fermentation media, cofactor recycling systems, immobilization carriers, and enzyme formulation excipients. The midstream involves enzyme screening, directed evolution, fermentation expression, purification, immobilization treatment, and process scale-up. Downstream applications are mainly concentrated in pharmaceutical biocatalysis, agrochemical intermediate, and specialty fine chemical. In pharmaceutical and CDMO applications, amine dehydrogenase can be used for the preparation of chiral amines, heterocyclic amines, aryl amine derivatives, and API intermediates. In agrochemical and specialty fine chemical applications, it can be used for the synthesis of amine-containing structural fragments, functional amine compounds, and customized high-value intermediates.
