Hedgehog Pathway Inhibitor Program

The Hedgehog pathway is normally active during embryonic development and regulates tissue and organ formation by directly promoting cell division in specific cell types, and by activating other secondary signaling pathways that control the synthesis of growth factors and angiogenic (blood vessel-forming) factors. The company’s Hedgehog pathway inhibitor technologies are being developed in various cancer indications under a collaboration agreement with Genentech, Inc., a wholly-owned member of the Roche Group.

The primary drug candidate being developed under this program is GDC-0449, an orally-administered small molecule Hedgehog pathway inhibitor. Genentech and Roche are responsible for the clinical development and commercialization of GDC-0449 and are is conducting three clinical trials of GDC-0449, including a pivotal phase II trial in advanced basal cell carcinoma that was initiated in 2009. Genentech is also conducting phase II clinical trials of GDC-0449 in colorectal and advanced ovarian cancer.

In addition to the ongoing clinical trials that Genentech and Roche are conducting, Genentech and the National Cancer Institute (NCI), entered into a collaborative relationship that allows the NCI to study GDC-0449 in additional potential cancer indications. Under this arrangement with NCI, third-party investigators began enrolling patients in a phase I clinical trial that is designed to evaluate dose and safety of GDC-0449 in young patients with medulloblastoma and a phase II trial to test the molecule in adult medulloblastoma patients.

Proprietary Targeted Cancer Programs

CUDC-101: CUDC-101 is the compound selected as a drug candidate from its targeted cancer programs. CUDC-101 is designed as a multi-target inhibitor of HDAC, EGFR, and Her2 and is the subject of a phase I clinical trial. In 2009, the company licensed its single-agent, single-target inhibitor drug candidate, Debio 0932, an Hsp90 inhibitor to Debiopharm.

In preclinical studies, CUDC-101 demonstrated the potential to inhibit all three molecular targets resulting in the potent killing of a range of cancer cell lines that are representative of various human cancer types, many of which have demonstrated resistance to various approved targeted agents. In 2008, the company initiated a phase I trial of CUDC-101 in patients with refractory solid tumors.

Debio 0932

In 2008, the company selected Debio 0932, an Hsp90 inhibitor, as a development candidate from its targeted cancer programs. In its preclinical studies, Debio 0932 demonstrated potent efficacy across a range of cancers in preclinical cancer models and exhibited promising pharmacological features in preclinical testing, particularly its high oral bioavailability, high tumor penetration and extended tumor retention.

In 2009, the company granted a worldwide license to develop, manufacture, market, and sell its Hsp90 inhibitor technology, including preclinical development candidate, Debio 0932, to Debiopharm.
