Rationale

The use of surface marker identification and classification of cells began with the revolution in immunology brought about by the discovery of T and B cells, which can be distinguished by virtue of specific surface markers. The technology has now spread to the subclassification of lymphocytes, and has begun to allow the identification of many other cell types, such as monocytes, macrophages, myeloid stem cells, and tumor cells.

The devices are now available to detect single cells that are capable of making antibody against a particular antigen, and to sort those cells into separate culture vessels. This permits the screening and cloning of cells in one operation, while automatically weeding out the dead cells in the process. A combination of the hybridoma technology with flow cytometry is synergistic, and will further expand the applications of both technologies.

During the next five years, the main trend in flow cytometry in general, and in cell surface marker analysis specifically, will be further simplification of the sample preparation and the analysis itself. Contains 120 pages and 15 tables
